- Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of U-87 MG (Lane 1), IMR-32 (Lane 2), Neuro-2a (Lane 3), SK-N-AS (Lane 4), T98G (Lane 5), tissue extract (30 µg lysate) of Rat Brain (Lane 6) and SH-SY5Y (Lane 7). The blot was probed with Anti-PGP9.5 Mouse Monoclonal Antibody (Product # MA5-12371, 1:250 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.4 µg/mL, 1:2500 dilution). A 25 kDa band corresponding to PGP9.5 was observed across the cell lines and tissue tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE and reg; 12% Bis-Tris gel (Product # NP0342BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with iBlot® 2 Dry Blotting System (Product # IB21001). The membrane was probed with the relevant primary and secondary Antibody follow+F32ing blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).

The blot was probed with anti-PGP9.5 Mouse Monoclonal Antibody (Product # MA5-12371,1:250 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4000 dilution). A 25 kDa band corresponding to PGP9.5 was observed in Mouse Brain, Rat Brain, U-87 MG, IMR32, Neuro-2a and SH-SY5Y and not observed in Rat Lung tissue which is documented to be PGP9.5 negative.

- Western blot analysis of PGP9.5/ UCHL1 was performed by loading 30 µg of SHSY5Y wild type (Lane 1), SHSY5Y Cas9 control (Lane 2), SHSY5Y PGP9.5/ UCHL1 knockout (Lane 3) whole cell extracts. The blot was probed with Anti-PGP9.5/ UCHL1 Monoclonal Antibody (Product # MA5-12371) (1:4000 dilution) and Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177) (1:4000 dilution). Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to PGP9.5/ UCHL1.
