Description

Bavituximab is a chimeric (mouse/human) monoclonal antibody that targets phosphatidylserine (PS). PS is a phospholipid normally confined to the inner leaflet of the plasma membrane in healthy cells but becomes externally exposed on the surface of cells undergoing apoptosis, and on tumor cells and virus-infected cells. Bavituximab was developed as a broad-spectrum anti-cancer and anti-viral therapeutic. Its proposed anti-tumor mechanism involves binding to exposed PS on tumor vascular endothelial cells and tumor cells, thereby blocking the immunosuppressive signals generated by PS and potentially promoting antibody-dependent cell-mediated cytotoxicity (ADCC). It has also been investigated for its antiviral effects by targeting PS on envelopes of viruses like hepatitis C virus (HCV) and SARS-CoV-2. Despite advancing to Phase III trials in combination with chemotherapy for non-small cell lung cancer, the trials did not meet their primary endpoints, and its development in oncology has been discontinued.
