- Western blot was performed using Anti-E-cadherin Monoclonal Antibody (HECD-1)(Product # 13-1700) and a 110kDa band corresponding to E-cadherin was observed across in all tested cell lines and tissues, except MDA-MB-231, SW480 and BJ. Whole cell extracts (30 µg lysate) of MCF7 (Lane 1), T-47D (Lane 2), MDA-MB-231 (Lane 3), SW480 (Lane 4), Caco-2 (Lane 5), BJ (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (5 µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). The additional bands at ~80kDa and 60kDa represent products of proteolytic cleavage of E-cadherin.

- Figure 2 Changes in cell morphology and gene expression under hypoxia. ( A ) Immunofluorescent staining (red) of the epithelial marker E-cadherin (HECD-1) and the mesenchymal marker vimentin (V9). Three human breast cell lines, MDA-MB-231, MCF-7, and MCF-10A cells were cultured in hypoxia for 72 h prior to staining. The secondary antibody is rabbit anti-mouse labelled with AlexaFluor 594. Cell nuclei were stained with DAPI. Relative fluorescence intensity of ( B ) E-Cadherin and ( C ) vimentin. Cells were cultured in normoxic and hypoxic conditions for 72 hours prior to the staining. Mean +- S.E.M. data are shown (n = 3); ** p < 0.01 (Student's t test). Scale bar is 100 um.
