Western blot was performed using Sortilin Polyclonal Antibody (Product # PA5-19481) and a 110kDa band corresponding to SORT1 was observed across all cell lines and tissues tested except Mouse and Rat liver. Whole cell extracts (30 µg lysate) of SK-BR-3 (Lane 1), MDA-MB-231 (Lane 2), BeWo (Lane 3), HeLa (Lane 4), Mouse brain (Lane 5), Rat brain (Lane 6), Mouse liver (Lane 7), Rat liver (Lane 8), and Mouse colon (Lane 9)were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
