Samples were incubated in TEK polyclonal antibody (Product # PA5-23082) using a dilution of 1.0 µg/mL followed by an anti-rabbit HRP secondary antibody. Separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. Detection: chemiluminescence.

- Western blot was performed using Anti-TEK Polyclonal Antibody (Product # PA5-23082) and a 150 kDa band corresponding to TEK was observed across cell lines and tissues tested except U-87 MG and Mouse Kidney which are reported to be negative. Whole cell extracts (30 µg lysate) of Hep G2 (Lane 1), MCF-7 (Lane 2), U-87 MG (Lane 3), HUVEC (Lane 4), HUVEC treated with Cobalt Chloride (75umol/ml for 24hr) (Lane 5), HUVEC treated with IL-1beta (1ng/ml, 24hr) (Lane 6), Mouse lung (Lane 7) and Mouse kidney (Lane 8) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (PA5-23082,1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005)..

- Immunohistochemical analysis of TEK in formalin-fixed paraffin-embedded tissue section of mouse stomach. Samples were incubated in TEK polyclonal antibody (Product # PA5-23082) using a dilution of 1:200 followed by HRP labeled anti-rabbit secondary antibody and DAB reagent. Nuclei of cells were counter-stained with hematoxylin.
