Western blot of ATF6 in NIH-3T3 cell lysate using an ATF6 monoclonal antibody (Product # MA1-25358) at a dilution of 3 µg/mL. A band corresponding to full-length ATF6 was detected. The ~36 kDa observed band may be an ATF6 breakdown/cleavage product.

Experimental details

Knockdown of ATF6 was achieved by transfecting HeLa cells with ATF6 specific siRNAs (Silencer® select Product # s22688, s223543). Western blot analysis (Fig. a) was performed using whole cell extracts from the ATF6 knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with ATF6 Monoclonal Antibody (Product # MA1-25358, 1:1000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25µg/ml, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to ATF6.

Experimental details

Western blot analysis was performed on whole cell extract (30 µg lysate) of MCF7 (Lane 1), MDA-MB-231 (Lane 2), A549 (Lane 3), U87MG (Lane 4), SHSY5Y (Lane 5), NIH3T3 (Lane 6) and HeLa (Lane 7). The blot was probed with Anti-ATF6 Monoclonal Antibody (70B1413) (Product # MA1-25358, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/ml, 1:4000 dilution). A 36 kDa band corresponding to the cleaved form of ATF6 was detected in all cell lines tested.
