- Western blot analysis was performed on whole cell extract (30 µg lysate) of Jurkat (Lane 1), MOLT-4 (Lane 2), SH-SY5Y (Lane 3) and tissue extract of Rat Brain (Lane 4). The blot was probed with Anti-CaMKIV Monoclonal Antibody (Product # MA5-17038, 1:500 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4000 dilution). A 52 kDa band corresponding to CaMKIV was observed across the cell lines and tissue tested.

- Knockdown of CaMKIV was achieved by transfecting SHSY5Y with CaMKIV specific siRNAs (Silencer® select Product # s2355). Western blot analysis (Fig. a) was performed using whole cell extracts from the CaMKIV knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with CaMKIV Monoclonal Antibody (8C5B8) (Product # MA5-17038, 1:1000 dilution) and Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Reduction of signal upon siRNA mediated knock down confirms that antibody is specific to CaMKIV.
