- Neutralization of FGF23 in NIH‚3T3 mouse embryonic fibroblast cell line. Samples were incubated in FGF23 polyclonal antibody (Product # PA5-46997). In the presence of Recombinant Mouse Klotho (5 µg/mL) and heparin (10 µg/mL), Recombinant Human FGF‚23 stimulates proliferation in the NIH‚3T3 mouse embryonic fibroblast cell line in a dose-dependent manner (orange line). Under these conditions, proliferation elicited by Recombinant Human FGF‚23 (1 µg/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human FGF‚23 Antigen Affinity-purified Polyclonal Antibody. The ND50 is typically 5‚15 µg/mL.

- Neutralization antibody testing demonstrates the specificity of an antibody through a correlation between antibody binding and the activity of the target. Neutralization of FGF23 is shown by the decrease in mean RFU (relative fluorescent units - measure of FGF-23-induced proliferation of NIH/3T3 cells) with increasing concentrations of FGF23 Polyclonal Antibody (PA5-46997).
