- Western blot analysis of IL17RA in K562 human chronic myelogenous leukemia cell line. Samples were incubated in IL17RA polyclonal antibody (Product # PA5-47199) using a dilution of 0.5 µg/mL followed by a HRP-conjugated Anti-Goat IgG secondary antibody. A specific band was detected for IL‚17 RA/IL‚17 R at approximately 160 kDa (as indicated). This experiment was conducted under reducing conditions.

- Neutralization antibody testing demonstrates the specificity of an antibody through a correlation between antibody binding and the activity of the target. Neutralization of IL-17RA is shown by the decrease in OD (IL-6 secretion by IL-17 stimulation is measured by IL-6 ELISA) with increasing concentrations of IL-17RA Antibody (Product # PA5-47199).

- Neutralization of IL17RA in normal human dermal fibroblasts. Samples were incubated in IL17RA polyclonal antibody (Product # PA5-47199). Recombinant Human IL‚17 stimulates IL‚6 secretion in normal human dermal fibroblasts in a dose-dependent manner (orange line), as measured by the Human IL‚6 Quantikine ELISA Kit. IL‚6 secretion elicited by Recombinant Human IL‚17 (25 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human IL‚17 RA/IL‚17 R Antigen Affinity-purified Polyclonal Antibody. The ND50 is typically 0.1-0.5 µg/mL.
