- Experimental details
- Knockdown of ELP3 was achieved by transfecting SH-SY5Y cells with ELP3 specific siRNA (Silencer® select Product # s30262 and Product # s30264). Western blot analysis (Fig a) was performed using whole cell extracts from the SH-SY5Y knockdown cells (Lane 3), non-specific scrambled siRNA transfected cells (Lane 2) and untransfected cells (Lane 1). The blots were probed with Anti-ELP3 Recombinant Rabbit Monoclonal Antibody (Product # 702669, 1:200 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution). Densitometric analysis of this Western blot is shown in histogram (Fig b). Loss of signal upon siRNA mediated knock down confirms that antibody is specific to ELP3.

- Experimental details
- Western blot analysis was performed on whole cell extracts (30 µg lysate) of IMR-32 (Lane 1), U-87 MG (Lane 2), HeLa (Lane 3), A549 (Lane 4), Neuro-2a (Lane 5), HT-29 (Lane 6), MDA-MB-231 (Lane 7) and tissue extract (30 µg lysate) of Mouse Brain (Lane 8). The blot was probed with Anti-ELP3 Recombinant Rabbit Monoclonal Antibody (Product # 702669, 1:200 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution). A ~62 kDa band corresponding to ELP3 was observed across the cell lines tested.
