Fixation: Samples can be stored in IC Fixation Buffer (cat. 00-8222) (100 µL cell sample + 100 µL IC Fixation Buffer) or 1-step Fix/Lyse Solution (cat. 00-5333) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency/compensation. Some generalizations regarding fluorophore performance after fixation can be made, but clone specific performance should be determined empirically.

Experimental details

Immunofluorescent analysis of ACO2 (orange) in HeLa cells. The cells were fixed with 4% Paraformaldehyde in PBS for 15 minutes at room temperature, and blocked with 3% BSA in PBS (Product # 37525) for 30 minutes at room temperature. Cells were stained with a ACO2 Monoclonal Antibody, AlexaFluor 555 conjugate (Product # MA1-029-A555) at a dilution of 1.25 µg/mL in blocking buffer for 1 hour at room temperature protected from light. Nuclei (blue) were stained with Hoechst Dye (Product # 62249) at a dilution of 1:10,000 in blocking buffer. Images were taken on a Thermo Scientific ToxInsight Instrument at 20X magnification.
