- Immunofluorescent analysis of HoxA5 (red) in HUES3::Hb9-GFP human embryonic stem cells patterned to cervical neuromesodermal progenitors as previously described (Lippmann et. al. 2015) and subsequently differentiated into motor neurons (green) using smoothened agonist, purmorphamine and retinoic acid. The cells were fixed with 4% paraformaldehyde for 10 minutes and blocked with 5% normal donkey serum in tris buffered saline containing 0.3% Triton-X (TBSDT) for at least one hour at 4C. Cells were stained with a monoclonal HoxA5 antibody (Product # PA5-50045; 1:100) and GFP antibody in TBSDT overnight at 4C, washed, and then incubated with an Alexa Fluor secondary antibodies (1:500) for one hour at room temperature. Nuclei (blue) were counterstained with DAPI. Images were taken on a Nikon AR1 confocal microscope at 40X magnification. Data courtesy of Dr Ashton's lab at University of Wisconsin at Madison.
