- Western blot analysis was performed on modified whole cell extracts (1% SDS) (30 µg lysate) of A-431 (Lane 1), A-431 treated with EGF (200 ng/mL for 10 mins) (Lane 2), A-431 treated with MK2206 (5uM for 5 hours) followed by EGF (200 ng/mL for 10 mins) (Lane 3), U-87 MG (Lane 4), U-87 MG treated with Wortmanin (2uM for 1 hr) (Lane 5), NIH/3T3 (Lane 6) and NIH/3T3 treated with PDGF (50 ng/mL for 30 mins) (Lane 7). The blot was probed with Anti-Phospho-c-Myc (Thr58) Polyclonal Antibody (Product # PA5-36673, 1:1000 dilution) and detected by chemiluminescence using Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). A 57 kDa band corresponding to Phospho-c-Myc (Thr58) was observed to increase upon EGF and PDGF treatment. Pre-treatment with AKT inhibitor (MK2206) and treatment with PI3K inhibitor (Wortmannin) resulted in inhibition of Phospho-c-Myc.
