- Western blot analysis of Methyl-PP2A alpha (Leu309) in two yeast strains using a Methyl-PP2A alpha (Leu309) monoclonal antibody (Product # MA5-18072). Native cultures from yeast deletion strains expressing the HA-tagged PP2AC (PPH21) were subjected to IP against the HA tag: strain PPE1-delta (left) which lacks the PP2A methylesterase (PPE1) and accumulates methylated PPH21, and PPM1-delta which lacks the PP2A methyltransferase and contains no methylated PPH21. The lysates were boiled in Laemmli buffer, separated by SDS-PAGE and transfered to a nitrocellulose membrane. Lane 5 shows a control incubated with an anti-HA tag monoclonal antibody.
