- Immunofluorescence analysis of ERBB4 was done on 70% confluent log phase A431 cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were subsequently labeled with ERBB4 Rabbit Polyclonal Antibody (Product # PA5-14637) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034) at a dilution of 1:1500 for 45 minutes at room temperature. Nuclei (Blue) were stained with SlowFade® Gold Antifade Mountant DAPI (S36938). F-actin (Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300). ERBB4 signal was observed in control cell line (panels a, d) and not in the ERBB4 knockout (KO) cell line (panels e, h). The images were captured at 60X magnification.

- Experimental details
- Immunohistochemistry analysis of ErbB4 in formalin-fixed and paraffin-embedded human cancer tissue. Samples were incubated with ErbB4 polyclonal antibody (Product # PA5-14637) which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of this antibody for immunohistochemistry; clinical relevance has not been evaluated. BC = breast carcinoma; HC = hepatocarcinoma.
