Western blot analysis of IGFBP2 using a IGFBP2 monoclonal antibody (Product # MA5-15400) against a truncated IGFBP2-His recombinant protein (1) and truncated IGFBP2 (aa40-328) human IgG Fc transfected CHO-K1 cell lysate (2).

Western blot was performed using Anti-IGFBP2 Monoclonal Antibody (1F6F6), (Product # MA5-15400) and a 35 kDa band corresponding to IGFBP2 was observed in the cell lines tested and increased upon protein transport inhibitor in SH-SY5Y. Membrane enriched cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), SH-SY5Y treated with Protein Transport Inhibitor (1X for 4 hours) (Lane 2), OVCAR-3 (Lane 3), and SK-OV-3 (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0321BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
