Experimental details

Western blot was performed using Anti-Alpha Synuclein Polyclonal Antibody (Product # PA1-18256) and a 15 kDa band corresponding to Alpha Synuclein was observed across positive tissue models Mouse and Rat Brain but not in negative models- Rat Heart and Liver, along with uncharacterized bands (*) at ~60 kDa and ~50 kDa. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Mouse Heart (Lane 2), Rat Brain (Lane 3), Rat Liver (Lane 4) and Rat Heart (Lane 5) were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1.25 µg/mL) and detected by chemiluminescence with Rabbit anti-Sheep IgG (H+L) Secondary Antibody, HRP (Product # 61-8620, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
