- Western blot analysis of EDG2 in different lysates. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. Samples were incubated in EDG2 Monoclonal antibody (Product # MA5-44840) using a dilution of 1:500 in 5% NFDM/TBST at room temperature for 2 hours followed by Goat Anti-Rabbit IgG - HRP secondary antibody at a dilution of 1:200,000 for 1 hour at room temperature. Positive control: Lane 1: Jurkat cell lysate; Lane 2: Hela cell lysate; Lane 3: A549 cell lysate.

- Immunohistochemistry analysis of EDG2 in paraffin-embedded human kidney tissue. The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with EDG2 Monoclonal antibody (Product # MA5-44840) using a dilution of 1:200 for 30 minutes at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

- Flow cytometry of EDG2 in HeLa cells. The cells were fixed, permeabilized and stained with EDG2 Monoclonal antibody (Product # MA5-44840) using a dilution of 1 µg/mL (red) at 4℃ for an hour followed by Alexa Fluor®488 conjugate-Goat anti-Rabbit IgG secondary antibody at a dilution of 1:1,000 for 30 minutes at 4℃ (dark incubation). Rabbit IgG Isotype Control (green). Unlabeled sample was used as a control (cells without incubation with primary antibody; black).
