Figure 1. Western blot using Affinity Purifiedanti-BACH1 antibody shows detection of a band at~105 kDa (lane 1) corresponding to human BACH1present in a 293 whole cell lysate (arrowhead). Lane 2shows that specific band staining is competed out whenthe antibody is pre-incubated with the peptideimmunogen prior to reaction. Approximately 35 µg oflysate was separated on a 4-20% Tris-Glycine gel bySDS-PAGE and transferred onto nitrocellulose. Afterblocking the membrane was probed with the primaryantibody diluted to 1:1,000. Reaction occurred 2 h atroom temperature followed by washes and reactionwith a 1:10,000 dilution of IRDye TM800 conjugated Gt-a-Rabbit IgG [H&L] MX for 45 min at roomtemperature (800 nm channel, green). Molecularweight estimation was made by comparison toprestained MW markers in lane M (700 nm channel,red). IRDye TM800 fluorescence image was capturedusing the Odyssey® Infrared Imaging Systemdeveloped by LI-COR. IRDye is a trademark of LI-COR,Inc. Other detection systems will yield similar results.
