- Western blot analysis of c-Ret was performed using membrane enriched extracts (30 µg lysate) of SH-SY5Y (Lane 1), SK-N-AS (Lane 2) and Neuro-2a (Lane 3). The blots were probed with Anti-c-Ret Mouse Monoclonal Antibody (Product # AHO1482, 2 µg/mL) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.4 µg/mL, 1:2500 dilution). A 170 kDa band corresponding to c-Ret was observed across the cell lines tested. Known quantity of protein samples were electrophoresed using Novex® NuPAGE® 10 % Bis-Tris gel (Product # NP0302BOX), XCell SureLock™ Electrophoresis System (Product # EI0002) and Novex® Sharp Pre-Stained Protein Standard (Product # LC5800). Resolved proteins were then transferred onto a nitrocellulose membrane with overnight wet transfer system. The membrane was probed with the relevant primary and secondary Antibody following blocking with 5 % skimmed milk. Chemiluminescent detection was performed using Pierce™ ECL Western Blotting Substrate (Product # 32106).

- Western blot was performed using Anti-RET Monoclonal Antibody (421R25) (Product # AHO1482) and 110-180kDa bands corresponding to RET were observed in SH-SY5Y, SK-N-AS, Neuro-2a and Rat Cerebellum. No bands were observed in HeLa, Hep G2, HEK-293, Rat Liver and Rat Kidney. Membrane enriched extracts (30 µg lysate) of SH-SY5Y (Lane 1), SK-N-AS (Lane 2), HeLa (Lane 3), Hep G2 (Lane 4), HEK-293 (Lane 5), Neuro-2a (Lane 6), Rat Cerebellum (Lane 7), Rat Liver (Lane 8) and Rat Kidney (Lane 9) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0321BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (2µg/mL) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752).
