- Western blot analysis of Degerative Spermatocyte Homolog 1 (DSH1) was performed by loading 40 µg of mouse retinal tissue lysates from 5 individual mice: 3 wild type mice (WT), where the photoreceptors are predominantly rods; and 2 mutant mice (Mutant), where the photoreceptors are predominantly cones per well onto an SDS-PAGE gel. Proteins were transferred to a nitrocellulose membrane and blocked with 5% milk in TBST for 1h RT. The membrane was probed with a DSH1 polyclonal antibody (Product # PA1-12912) at a dilution of 1:1000 4°C, washed, and probed with an HRP-conjugated anti-rabbit IgG secondary antibody at a dilution of 1:10,000 for 1 hour at room temperature. - Immunohistochemistry was performed on 4% paraformaldehyde-fixed frozen mouse retina tissue sections. Tissues were blocked in 3% normal goat serum + 0.3% Triton X-100 for 1 hour at room temperature and probed with a Degenerative Spermatocyte Homolog 1 (DSH1) polyclonal antibody (Product # PA1-12912) at a dilution of 1:500 overnight at 4°C. Tissues were washed extensively with PBS. Detection was performed using a Cy3-conjugated goat anti-rabbit IgG secondary antibody at a dilution of 1:500.
