- AbFlex Histone H4K5ac/K8ac recombinant antibody (rAb) tested by Western blot 20 µg of HeLa cell nuclear extract was run on SDS-PAGE and probed with AbFlex Histone H4K5ac/K8ac antibody at 2 µg/mL. Lane 1: nuclear extract from untreated cells. Lane 2: nuclear extract from cells treated with sodium butyrate.

- AbFlex Histone H4K5ac/K8ac recombinant antibody (rAb) specificity tested by ELISA 10, 2 or 0.4 pmole of various biotinylated Histone H4 peptides were captured in duplicate wells of a Streptavidin coated plates. 0.5 µg/mL of Histone H4K5ac/K8ac antibody was added and detected using secondary anti-mouse HRP antibody. Histone H4K5ac/K8ac antibody recognizes peptide containing both K5ac and K8ac. The presence of additional K12 and K16 acetylation does not affect detection.

- AbFlex Histone H4K5ac/K8ac recombinant antibody (rAb) tested by ChIP-Seq Chromatin immunoprecipitation (ChIP) was performed using the ChIP-IT High Sensitivity Kit (Cat. No. 53040) with 30 µg of HeLa cell chromatin and 4 µg of antibody. ChIP DNA was sequenced on the Illumina NextSeq and 6.7 million sequence tags were mapped to identify Histone H4K5ac/K8ac binding sites on chromosome 1.
