- Detection of Human HSP70/HSPA1A by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line untreated (-) or treated (+) with 42˚ C heat shock for 30 minutes and allowed to recover for 3 hours. PVDF membrane was probed with 2 µg/mL of Mouse Anti-Human/Mouse/Rat HSP70/HSPA1A Monoclonal Antibody (Catalog # MAB16631) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF018). A specific band was detected for HSP70/HSPA1A at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Simple Western lane view shows lysates of Jurkat human acute T cell leukemia cell line untreated (-) or treated (+) with 42oC heat shocked (HS) for 30 minutes and allowed to recover for 3 hours, loaded at 0.2 mg/mL. A specific band was detected for HSP70/HSPA1A at approximately 70 kDa (as indicated) using 20 µg/mL of Mouse Anti-Human HSP70/HSPA1A Monoclonal Antibody (Catalog # MAB16631) . This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.

- HSP70/HSPA1A in Human Kidney Cancer Tissue. HSP70/HSPA1A was detected in immersion fixed paraffin-embedded sections of human kidney cancer tissue using Mouse Anti-Human/Mouse/Rat HSP70/HSPA1A Monoclonal Antibody (Catalog # MAB16631) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell nuclei.
