- Description: The monoclonal antibody MFCDA84 recognizes mouse and human Undifferentiated embryonic cell Transcription Factor 1 (UTF1) which was first identified in mouse embryonic stem cells (inner cell mass and the epiblast) but is also expressed in human embryonic and adult stem cells. UTF1 transcription ceases at the onset of differentiation, which distinguishes it from less-sensitive pluripotency markers, such as Oct3/4 or Nanog. Importantly, its transcriptional regulation is very sensitive to differentiation cues and is switched off faster and with a larger magnitude than either Oct3/4 or Sox2 during embryoid body formation from both mouse and human ESCs. Reduction in UTF1 expression is associated with differentiation, although it does not appear to play a direct role in self renewal. UTF1 expression is regulated by Oct3/4 and is associated with chromatin. It is a transcriptional co-activator of ATF2 and is thought to play a role in maintaining an epigenetic environment necessary for maintained pluripotency. Co-expression of UTF1 with the reprogramming factors c-Myc, Oct3/4, Sox2, and KLF4, as well as reduction of p53 increases efficiency of iPS cell generation by 100 fold. In addition to its expression in stem cells, UTF1 is expressed in neuroblastoma and the presence of UTF1 along with BMI1 can distinguish low grade neuroblastomas from stage IV tumors. Applications Reported: This MFCDA84 antibody has been reported for use in western blotting, immunohistochemical staining of formalin-fixed paraffin embedded tissue, ELISA, and immunocytochemistry. Applications Tested: This MFCDA84 antibody has been tested by immunocytochemistry on fixed and permeabilized cells, and immunohistochemistry on FFPE human tissue using both high and low pH antigen retrieval and can be used at less than or equal to 10 ug/mL. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest. Purity: Greater than 90%, as determined by SDS-PAGE. Aggregation: Less than 10%, as determined by HPLC. Filtration: 0.2 µm post-manufacturing filtered.
