- Immunohistochemistry analysis of Fibroblast Growth Factor (FGF2) in the equine placenta. Tissue was formalin fixed and paraffin embedded. Staining was performed in Bond-MAX automated system following manufacturer’s protocols. Briefly, automated dewaxing and rehydration step, followed by 100°C for 10 min antigen retrieval using an EDTA based retrieval solution (pH of 8.2). Post-retrieval, slides were incubated with 3% hydrogen peroxide (5 minutes), diluted primary antibody (30 minutes), blocking reagent (8 minutes), horseradish peroxidase-labeled polymer (8 minutes), and diaminobenzidine substrate (10 minutes). Primary antibody FGF2 (Product # OSG00014W) was diluted 1:200. Counterstain was hematoxylin. Sections were dehydrated and mounted with a coverslip. Images were taken at 100X magnification. Notably, FGF2 immunohistochemistry analysis produced heavy tissue staining. The negative control was assessed (omission of primary antibody), and no staining was observed (data not shown).
