- Experimental details
- Western blot was performed using Anti-BDNF Polyclonal Antibody (Product # PA1-18369) and a 34 kDa band corresponding to Brain-derived neurotrophic factor was observed across the panel tested except for Mouse liver and spleen which are reported to be negative. Tissue extracts (30 µg lysate) of Mouse Brain (Lane 1), Mouse Liver (Lane 2), Mouse Spleen (Lane 3), Rat Brain (Lane 4) and Rat Spleen (Lane 5) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (5 µg/mL) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Reagent Kit (Product # WP20005). An uncharacterized band (*) of ~40 kDa was also observed in mouse and rat brain tissues.
