Description
Fc receptor-like protein 5 is encoded by the FCRL5 gene in human. FcRH5 is a single-pass type I membrane protein that is expressed in marginal zone B-cells, immunoblasts, tonsillar germinal center centrocytes and in the intraepithelial and interfollicular regions of the tonsil. It is involved in B-cell development and differentiation in peripheral lymphoid organs and may be useful markers of B-cell stages. It is shown to regulate BCR signaling and binds aggregated IgG and engages several of its Ig-domains to bind IgG. It is shown to be overexpressed in many lymphoma cell lines and on hairy cell leukemia cells. Serum levels of soluble FcRH5 are reported to be elevated in patients with several types of B cell tumors. FcRH5 is synthesized with a signal peptide (aa 1-15), which is cleaved off in the mature form that has a long extracellular domain (aa 16-851), a short transmembrane domain (aa 852-872), and a cytoplasmic tail (aa 873-977) that contains immunoreceptor tyrosine-based inhibitor motifs (ITIM).
