Experimental details

Western blot analysis was performed on membrane enriched extracts (30 µg lysate) of SK-BR-3 (Lane 1), T-47D (Lane 2) and MDA-MB-231 (Lane 3). The blots were probed with Anti-ErbB2 Sheep Polyclonal Antibody (Product # PA1-12361, 5 µg/mL) and detected by chemiluminescence using Rabbit anti-Sheep IgG (H+L) Secondary Antibody, HRP conjugate (Product # 61-8620, 0.25 µg/mL, 1:4000 dilution). A 185 kDa band corresponding to ErbB2 was observed in SK-BR-3 and T-47D but not in MDA-MB-231, which is a negative control for ErbB2.
