- Western Blot: Histone H4 [Methyl Lys20] Antibody (C.15100147) [NBP2-59234] - Histone extracts of HeLa cells (15 ug) were analyzed by Western blot using the antibody against H4K20me1 at 1:1000 in TBS-Tween containing 5% skimmed milk. The position of the protein of interest is indicated on the right; the marker (kDa) is shown on the left.

- Chromatin Immunoprecipitation: Histone H4 [Methyl Lys20] Antibody (C.15100147) [NBP2-59234] - ChIP assays were performed using human HeLa cells, the antibody against H4K20me1 and optimized PCR primer sets for qPCR. ChIP was performed with a LowCell number ChIP kit, using sheared chromatin from 10,000 cells. Two different quantities of antibody (3 and 12 uL per ChIP experiment) were analyzed. IgG (1 ug) was used as negative IP control. QPCR was performed with primers for the GAPDH promoter and for the inactive gene TSH2B. Figure shows the recovery, expressed as % of input (the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).

- Dot Blot: Histone H4 [Methyl Lys20] Antibody (C.15100147) [NBP2-59234] - To check the specificity of the antibody against H4K20me1 a Dot Blot was performed with peptides containing different modifications or unmodified sequences of histone H4. 100 to 0.2 pmol of peptide containing the respective histone modification were spotted on a membrane. The antibody was used at a dilution of 1:10,000. Figure shows a high specificity of the antibody for the modification of interest.
