- Western Blot: Histone H4 [ac Lys8] Antibody (C.15200217) [NBP2-59240] - Histone extracts (15 ug) from HeLa cells were analyzed using the antibody against H4K8ac diluted 1:1,000 in TBS-Tween containing 5% skimmed milk. - Chromatin Immunoprecipitation: Histone H4 [ac Lys8] Antibody (C.15200217) [NBP2-59240] - Analysis was performed using HeLa cells, the monoclonal antibody against H4K8ac and optimized PCR primer sets for qPCR. ChIP was performed with a ChIP-seq kit, using sheared chromatin from 1.5 million cells. A titration of the antibody consisting of 0.5, 1, 2 and 5 ug per ChIP experiment was analysed. IgG (1 ug/IP) was used as negative IP control. QPCR was performed with primers for the promoter of the EIF4A2 gene, used as a positive control, and for the Sat2 satellite repeat, used as a negative control. Figure shows the recovery, expressed as a % of input (the relative amount of immunoprecipitated DNA compared to input DNA after qPCR analysis).

- Dot Blot: Histone H4 [ac Lys8] Antibody (C.15200217) [NBP2-59240] - To test the cross reactivity of the antibody against H4K8ac, analysis was performed with peptides containing other acetylations and unmodified sequences of histone H4. 100 to 0.2 pmol of the respective peptides were spotted on a membrane. The antibody was used at a dilution of 1:20,000. Figure shows a high specificity of the antibody for the modification of interest.
