- Immunohistochemical analysis of IL-33 in formalin-fixed paraffin-embedded tissue sections of human tonsil probed with ACD RNAScope® Probe and stained using ACD RNAscope® 2.5 HD Detection Reagents-Red (top image). Samples were incubated in IL-33 polyclonal antibody (Product # PA5-47006) using a dilution of 3 µg/mL with overnight incubation at 4 °C followed by Anti-Goat IgG VisUCyte HRP Polymer Antibody and DAB chromogen (lower image, yellow-brown). Tissues were counterstained with hematoxylin (blue).

- Immunohistochemical analysis of IL-33 in immersion fixed paraffin-embedded sections of human tonsil. Samples were incubated in IL-33 polyclonal antibody (Product # PA5-47006) using a dilution of 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown) and counterstained with hematoxylin (blue). Specific staining was localized to nuclei.

- Neutralization antibody testing demonstrates the specificity of an antibody through a correlation between antibody binding and the activity of the target. Neutralization of IL-33 is shown by the decrease in mean RFU (relative fluorescent units - measure of IL-33-induced proliferation of D10.G4.1 mouse helper T cell line) with increasing concentrations of IL-33 Polyclonal Antibody (PA5-47006).
