- Western blot was performed using Anti-CTGF Polyclonal Antibody (Product # PA1-22376) and a 38kDa band corresponding to Connective tissue growth factor was observed across in HUVEC upon PTI treatment, but not in HEK 293 upon PTI treatment. Whole cell extracts (30 µg lysate) of HUVEC (Lane 1), HUVEC (Protein Transport Inhibitor Cocktail- 500X (1X, 4 hours) (Lane 2), HEK-293 (Lane 3), HEK-293 (Protein Transport Inhibitor Cocktail- 500X (1X, 4 hours) (Lane 4) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0301BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:5000) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using SuperSignal™ West Dura Extended Duration Substrate (Product # 34076).

- Figure 3 Pro-fibrotic gene expression in TGF-beta induced fibroblasts. nCF transfected with scr siRNA or Fibulin-6 siRNA, after being exposed to TGF-beta were subjected to RNA or protein extraction for real-time PCR or western blotting respectively. ( a ) Collagen I alpha1 and ( b ) CTGF mRNA expression is increased after TGF-beta stimulation in scr- but not in fibulin-6-siRNA transfected nCF (n = 7, p < 0.05, nonparametric Mann-Whitney U test). Real-time RT-PCR signals were normalized to hypoxanthine phosphoribosyl-transferase (HPRT) gene expression and data are expressed as 2 -DeltaDeltaCT . ( c and d ) Densitometric analysis of western blots revealed upregulation of ( c ) collagen I alpha1 and ( d ) CTGF upon TGF-beta stimulation in scr-siRNA transfected nCF. In fibulin-KD nCF no upregulation could be observed (n = 5, p < 0.05, nonparametric Mann-Whitney U test).
