- Western blot analysis of PKC Pan in Lane 1: the Spleen tissue lysate of mouse (40 µg), Lane 2: the Brain tissue lysate of rat (40 µg), Lane 3: HeLa whole cell lysate (20 µg), Lane 4: HEK293T whole cell lysate (40 µg), Lane 5: A549 whole cell lysate (40 µg). Samples were incubated with PKC Pan polyclonal antibody (Product # PA5-36757) at a dilution of 1:500.

- Knockdown of PKC Pan was achieved by transfecting NIH/3T3 cells with PKC Pan specific siRNAs (PKC A, B, E, I and Q) (Silencer® select Product # s11094, s11092, s11097, s11096, s11102, s11101, s11110, s11122, s11123). Western blot analysis (Fig. a) was performed using whole cell extracts from PKC Pan knockdown cells (lane 3), non-specific scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blots were probed with PKC Pan Polyclonal Antibody (Product # PA5-36757, 1:1000 dilution) and Goat anti-Rabbit IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A27036, 0.25 µg/mL, 1:4000 dilution). Densitometric analysis of this western blot is shown in histogram (Fig. b). Reduction of signal upon siRNA mediated knock down confirms that antibody is specific to PKC Pan.
