- Western Blot staining of Human Peripheral Blood Mononucleocytes lysate using Product # PA5-19075 at a concentration of 0.5 µg/mL, the primary antibody incubation was 1 hour and the detection method was chemiluminescence.

- Western blot was performed using Anti-S100A9 Polyclonal Antibody (Product # PA5-19075) and a 13kDa band corresponding to S100A9 was observed in THP-1 but was absent in Raji and SH-SY5Y which are reported to be negative. Expression of S100A9 was downregulated upon treatment of THP-1 with PMA/TPA [10.1073/pnas.0709958105]. Membrane enriched cell extracts (30 µg lysate) of THP-1 (Lane 1), THP-1 treated with PMA/TPA (50ng/ml for 24 hours) (Lane 2), Raji (Lane 3) and SH-SY5Y (Lane 4) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1µg/ml dilution) and detected by chemiluminescence Rabbit Anti-Goat IgG Secondary Antibody, HRP conjugate (Product # A27014, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005). An uncharacterized band (*) was observed at ~50kDa in the negative cell models.

- Flow cytometric analysis of S100A9 in MCF7 cells using a polyclonal antibody (Product #PA5-19075). MCF7 cells (blue line) were paraformaldehyde fixed and permeabilized with 0.5% Triton. The primary antibody was incubated for one hour (10 µg/mL) followed by an Alexa Fluor 488 secondary antibody (1 µg/mL). IgG control: Unimmunized goat IgG (black line) followed by an Alexa Fluor 488 secondary antibody.
