- Western blot analysis of FZR1 in A431 whole cell and nuclear extracts using FZR1 polyclonal antibody (Product # PA5-85303) using 30 µg of sample at a dilution of 1:1000. Prior to incubation with primary antibody, the sample was separated on 10% SDS-PAGE.

- Knockdown of Fizzy-related protein homolog was achieved by transfecting HeLa with Fizzy-related protein homolog specific siRNAs (Silencer® select Product # s27991, s27992). Western blot analysis (Fig. a) was performed using Nuclear enriched extracts from the Fizzy-related protein homolog knockdown cells (lane 3), non-targeting scrambled siRNA transfected cells (lane 2) and untransfected cells (lane 1). The blot was probed with FZR1 Polyclonal Antibody (Product # PA5-85303, 1:1500) and Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000). Densitometric analysis of this western blot is shown in histogram (Fig. b). Decrease in signal upon siRNA mediated knock down confirms that antibody is specific to Fizzy-related protein homolog.

- Western blot was performed using Anti-FZR1 Polyclonal Antibody (Product # PA5-85303) and a 55kDa band corresponding to Fizzy-related protein homolog was observed. Nuclear enriched extracts (30 µg lysate) of HeLa (Lane 1), T98G (Lane 2), T-47D (Lane 3), HCT 116 (Lane 4), Jurkat (Lane 5) and tissue extract (30 µg lysate) of Mouse Testis (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1500) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
