- Western blot analysis was performed on nuclear extracts (30 µg lysate) of HCT-116 (Lane 1), HCT-116 treated with Paclitaxel (100 nM, 18 hrs)(Lane 2), HT-29 (Lane 3), HT-29 treated with Paclitaxel (100 nM, 18 hrs) (Lane 4), SK-BR-3 (Lane 5), SK-BR-3 treated with Nocodazole (100 ng/mL, 16 hrs) (Lane 6). The blot was probed with Aurora A Monoclonal Antibody (4G10)(Product #MA51-5804, 1:2,000 dilution) and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, HRP conjugate (Product # A28177, 0.25 µg/mL, 1:4,000 dilution). A 48 kDa band corresponding to Aurora A was observed upon the indicated treatments in the cell lines. The bands marked with * at 40 kDa and 35k Da may represent Aurora B and C respectively, however, their identity could not be confirmed.
