Western Blot: Raf1 [p Ser259] Antibody [NSB505] - Hek293 cell lysates overexpressing c-Raf alone (1) or immunoprecipitates prepared from cells cotransfected with c-Raf, Ras[V12] and Rac1 (2-6) were resolved by SDS-PAGE on 10% polyacrylamide gel and transferred to PVDF, then blocked with 5% BSA-TBST buffer ON at 4C, then incubated with the c-Raf [pSpY338/340] Ab for 2hrs at RT in a 3% BSA-TBST buffer, following prior incubation with: no peptide (1, 2), non-phosphopeptide corresponding to the phosphopeptide immunogen (3), generic phosphoserine-containing peptide (4), generic phosphotyrosine-containing peptide (5), or the phosphopeptide immunogen (6). Data show that only the peptide corresponding to c-Raf [pSpY338/340] blocks the Ab signal, demonstrating the specificity of the Ab.
