- 5 Figure Evidence of CatSper channel functionality and intracellular calcium. (a) Western blot analysis of sperm lysates indicates a high protein level of CatSper-2 proteins in the sperm cryopreserved in LDL extenders. (b,c) A relative quantification of CatSper-1 and CatSper-2 in a cryopreserved sperm. (d) Intracellular calcium estimation by flow cytometry using Fluo-3 AM/PI. Sperm gating was performed using a logarithm of side and forward scatter. A total of 10,000 events were examined for each sample. The gated sperm divided was into four windows--lower left (a): live sperm with low intracellular calcium; lower right (b): live sperm with high intracellular calcium; upper left (c): dead sperm with low intracellular calcium; upper right (d): dead sperm with high intracellular calcium. (e) The percentage of the live sperm with low intracellular calcium, live sperm with high intracellular calcium, dead sperm with low intracellular calcium, and dead sperm with high intracellular calcium.
