HeLa whole cell and nuclear extracts (30 µg) were separated by 10% SDS-PAGE, and the membrane was blotted with TEF1 Monoclonal Antibody (GT13112) (Product # MA5-27786) diluted at 1:500. The HRP-conjugated anti-mouse IgG antibody was used to detect the primary antibody.

- Western blot was performed using TEF1 Monoclonal Antibody (GT13112) (Product # MA5-27786) and a~48 kDa band corresponding to TEAD1 was observed across all the cell lines tested along with an uncharacterized band (*) at 30 kDa. Whole cell extracts (30 µg lysate) of A431 (Lane 1), HeLa (Lane 2), U2OS (Lane 3), BeWo (Lane 4), HEL 92.1.7 (Lane 5), U937 (Lane 6) and MOLT4 (Lane 7) were electrophoresed using NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001).The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).

Green: TEF1 protein stained by TEF1 Monoclonal Antibody (GT13112) (Product # MA5-27786) diluted at 1:400. Red: Phalloidin, a F-actin marker, diluted at 1:200.
