- Flow cytometry of BMAL1 in HeLa cells. Samples were incubated in BMAL1 polyclonal antibody (Product # PA1-46118) using a dilution of 5 µg/mL for 30 minutes at room temperature. Antibody (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Both antibodies were conjugated to Alexa Fluor 647.

- Flow cytometry of BMAL1 in Jurkat cells and a matched isotype control. Samples were incubated in BMAL1 polyclonal antibody (Product # PA1-46118) using a dilution of 2.5 µg/mL for 30 minutes at room temperature followed by a Rabbit IgG (H+L) Cross-Adsorbed secondary antibody. Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin.

- Fig. 3 Immunohistochemical analyses of Bmal1 and GFAP staining in SCN of Tg2576 (TG) and wild-type (WT) mice. A) Representative confocal images of Bmal1 and GFAP stained sections through SCN of 18-month-old WT and TG mice. Scale bar, 250mum (first and third column); 60mum (second and fourth column) and 20mum (in magnifications). B) Quantitative analysis of Bmal1 and GFAP staining in SCN of 18 months old WT and Tg2576 (TG) mice ( n = 6 per group). ** p < 0.01, * p < 0.05. GFAP, glial fibrillary acidic protein.
