- Detection of April in Human Blood Monocytes by Flow Cytometry. Human peripheral blood monocytes stimulated with 5 ng/mL Recombinant Human IL-4 (Catalog # 204-IL) and 10 μg/mL Goat Anti-Human IFN-gamma Antigen Affinity-purified Polyclonal Antibody (Catalog # AF-285-NA) for three days followed by a 3 hour incubation with monensin were stained with Mouse Anti-Human APRIL/TNFSF13 Monoclonal Antibody (Catalog # MAB8843) followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0102B) and Mouse Anti-Human CD4 APC-conjugated Monoclonal Antibody (Catalog # FAB3791A). Quadrant markers were set based on control antibody staining (Catalog # MAB004). To facilitate intracellular staining, cells were fixed with paraformaldehyde and permeabilized with saponin.
