Western blot was performed using Anti-MYH3 Polyclonal Antibody (Product # PA5-54514) and a 230 kDa band corresponding to MYH3 was observed across tissues tested except in Mouse Ovary and Testis and Rat Testis which are reported to be negative. Tissue extracts (30 µg lysate) of Mouse Heart (Lane 1), Mouse Skeletal Muscle (Lane 2), Mouse Testis (Lane 3), Mouse Ovary (Lane 4), Rat Heart (Lane 5) and Rat Testis (Lane 6) were electrophoresed using NuPAGE™ 3-8% Tris-Acetate Protein Gel (Product # EA0378BOX). Resolved proteins were then transferred onto a Nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
