- Figure 2 Differential effect of pathway-selective NTM peptides in cultured cells. ( A-C ) Treatment with Imp alpha5-selective NTM, cSN50.1alpha inhibits nuclear import of SRTFs: NF-kappaB RelA ( A ), phosphorylated STAT1 ( B ), and component of AP-1 complex, cFos ( C ) in LPS-stimulated RAW cells. Cells were pretreated with 30 uM NTM peptides (biselective cSN50.1, and two monoselective cSN50.1alpha and cSN50.1beta) for 30 min then incubated with LPS (10 ng/mL) for 6 h. ( D ) Nuclear import of nSREBP2 is inhibited by treatment with Imp beta1-selective NTM, cSN50.1beta peptide, in sterol-depleted HEK 293T cells. Cells were rapidly depleted of sterols by 15 min incubation with hydroxypropyl-beta-cyclodextrin (HPCD) and treated with NTM peptides (as indicated) for 2 h.
