- Western blot analysis of Enpp2 from zebrafish embryo lysate by Enpp2 polyclonal antibody ( Cat # PAB8520 ) (1:500, 4°C, overnight). Instead of a predicted 93.6 KDa band, we detected a ~50KDa single band from shield stage of wild type embryos (lane 1) on a 12% SDS-PAGE, and this band can be efficiently knockdown by Enpp2 morpholino oligonucleotides (land 2 and 3). This phenomenon is probably due to a cleavage of endogenous Enpp2 under reducing condition of SDS-PAGE as previously reported in human and mice.
