- Immunofluorescence analysis of SIAH1 was performed using 70% confluent log phase SW480 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 15 minutes, and blocked with 2% BSA for 1 hour at room temperature. The cells were labeled with SIAH1 Rabbit Recombinant Monoclonal Antibody (PSH0-46) (Product # MA5-49361) at 1:100 dilution in 0.1% BSA, incubated at 4°C overnight and then labeled with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A27034), (1:2,000 dilution), for 45 minutes at room temperature (Panel a: Green). Nuclei (Panel b: Blue) were stained with ProLong™ Diamond Antifade Mountant with DAPI (Product # P36962). F-actin (Panel c: Red) was stained with Rhodamine Phalloidin (Product # R415, 1:300 dilution). Panel d represents the merged image showing cytoplasmic localization. Panel e represents control cells with no primary antibody to assess background. The images were captured at 60X magnification.

- Immunofluorescence analysis of SIAH1 using A549 cells. The cells were fixed 4% paraformaldehyde for 10 minutes, permeabilized with 0.05% Triton™ X-100 in PBS for 20 minutes, and blocked with 2% negative goat serum for 30 minutes at room temperature. The cells were labeled with SIAH1 Rabbit Recombinant Monoclonal Antibody (PSH0-46) (Product # MA5-49361) at 1:100 dilutionin 2% negative goat serum overnight at 4°C and then with iFluor™ 488 Goat Anti-Rabbit IgG H&L secondary antibody (1:1,000 dilution) for 1 hour at room temperature (green). Nuclear were stained with DAPI (blue). Beta tubulin (red) was stained at 1:200 dilution overnight at +4°C. iFluor™ 594 Goat Anti-Mouse IgG H&L was used as the secondary antibody at 1:1,000 dilution. The images were captured at 200X magnification.

- Flow Cytometry analysis of SIAH1 in A549 cells. The cells were fixed and permeabilized and then stained with SIAH1 Rabbit Recombinant Monoclonal Antibody (PSH0-46) (Product # MA5-49361) at 1 µg/mL (red) and Rabbit IgG Isotype Control (green). After incubation of the primary antibody at 4°C for an hour, the cells were stained with a iFluor™ 488 Goat anti-Rabbit IgG Secondary antibody (HA1121) at 1:1,000 dilution for 30 minutes at 4°C.
