- Immunofluorescence analysis of MUC4/Mucin 4 was done on 70% confluent log phase MCF-7 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton™ X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with MUC4/Mucin 4 (1G8) Mouse Monoclonal Antibody (Product # 35-4900) at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Mouse IgG (H+L) Superclonal™ Secondary Antibody, Alexa Fluor® 488 conjugate (Product # A28175) at a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with SlowFade® Gold Antifade Mountant with DAPI (Product # S36938). F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin (Product # R415, 1:300). Panel d is a merged image showing cytoplasmic localization. Panel e is a no primary antibody control. The images were captured at 60X magnification.

- Figure 5 Quantitativeexpression levels for CLDN4, MSLN, MUC4, and CDH11on Panc-1, MIA PaCa-2, Capan-1, and HPDE cells. (A) phase and fluorescenceimages of MiaPaCa-2, Panc-1, Capan-1, and HPDE cells incubated withQD-FcR-Ab conjugates (Ab = aCLDN4, aMSLN, aMUC4, and aCDH11). Biomarkerexpression levels on (B) MIA PaCa-2, (C) Panc-1, (D) Capan-1, and(E) HPDE cells. Error bars represent standard error. Expression levelsfor each biomarker on MIA PaCa-2, Panc-1, and HPDE are obtained frommeasurements of between 565-997 individual cells (see Supporting Information Figure S2 for details). For Capan-1, expression levels are based on measurements of at least180 clusters of cells. In total, measurements were made on more than9600 individual cells and more than 780 cell clusters.
