- Reconstitute in 100 µL of sterile water. Centrifuge to remove any insoluble material. After reconstitution keep aliquots at -20 °C for a higher stability, and at 4 °C with an appropriate antibacterial agent. Glycerol (1:1) may be added for an additional stability. Avoid repetitive freeze/thaw cycles.

- Western blot analysis (Fig a) of alpha Synuclein was performed on whole cell extracts (30µg of lysate) of HeLa wild type (Lane 1) and alpha Synuclein knockout (Lane 2). The blot was probed with anti-alpha Synuclein Polyclonal Antibody (Product # PA1-18264, 1:2000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Loss of signal upon CRISPR mediated knockout (KO) confirms that antibody is specific to alpha Synuclein.

- Western blot was performed using Anti-Alpha Synuclein Polyclonal Antibody (Product # PA1-18264) and a 15 kDa band corresponding to Alpha Synuclein was observed across positive cell lines (SH-SY5Y and HEK-293,(Mouse Brain and Rat Brain) but not in negative models (Hep G2, A549, Mouse Heart, Rat Liver and Rat Heart). Whole cell extracts (30 µg lysate) of SH-SY5Y (Lane 1), HEK-293 (Lane 2), Hep G2 (Lane 3), A549 (Lane 4), tissue extracts of Mouse Brain (Lane 5), Mouse Heart (Lane 6), Rat Brain (Lane 7), Rat Liver (Lane 8) and Rat Heart (Lane 9) were electrophoresed using Novex® NuPAGE® 12 % Bis-Tris gel (Product # NP0342BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L), Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
