- Western blot was performed using PAX7 Polyclonal Antibody (Product # PA5-23990) and a 55 kDa band corresponding to PAX7 was observed in cell lines and tissue tested. Whole cell extracts (30 µg lysate) of A-673 (Lane 1), RD (Lane 2), C2C12 (Lane 3), C2C12 differentiated to myotubes (Lane 4), Mouse Heart (Lane 5), Mouse Liver (Lane 6) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23002) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:1000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:20,000 dilution) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580). PAX7 is highly expressed in A673, RD; C2C12 when compared to undifferentiated C2C12 and mouse heart when compared to mouse liver. Additional uncharacterized bands were observed across samples tested.
