- Western blot was performed using Anti-FAAH Rabbit Polyclonal Antibody (Product # PA5-52433) and a 60kDa band corresponding to FAAH was observed in LNCaP, MCF7, Mouse Brain, Mouse Liver, Rat Liver and not in PC-3, DU 145 which are reported negative for FAAH expression. Whole cell extracts of LNCaP (Lane 1), PC-3 (Lane 2), DU 145 (Lane 3), MCF7 (Lane 4), Mouse Brain (Lane 5), Mouse Liver (Lane 6) amd Rat Liver (Lane 7) were electrophoresed using Novex® NuPAGE® 4-12 % Bis-Tris gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:500 dilution) and detected by chemiluminescence Goat Anti-Rabbit IgG Secondary Antibody, HRP conjugate (Product # A27036, 1:4000 dilution) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
