- 96 well plates were pre-coated with 5 µg/mL rabbit anti-human GPT polyclonal antibody (Product # PA5-27453, blue bars) or control antibody (red bars) at 4 C overnight. Sandwich ELISA analysis of human GPT was performed by loading 100 ul per well of GPT recombinant protein at the concentration of 3000,1500, 750, 375, 187.5, 93.75, 46.875 and 0 ng/mL in 1% BSA-PBS. After for 1 hour incubation at room temperature with shaker, the plate was washed and then incubated with 50 ul per well of a biotinylated rabbit anti-human GPT polyclonal antibody (Product # PA5-14376, dilution 1:100) plus 50 ul per well of a biotinylated rabbit anti-human GPT polyclonal antibody (Product # PA5-14377 , dilution 1:100) for 1 hour at room temperature with shaker. The plate was washed and incubated with 100 ul per well of Streptavidin-HRP in PBS in all test wells for 1 hour at room temperature. Detection was performed using TMB Substrate for 20 minutes at room temperature in the dark. The plate was then stopped with 2N H2SO4. Absorbances were read on a spectrophotometer at 450 nm.
